I have a Mitel 3300 system, and am having a problem switching a newly added DN from single line to a multicall Prime line type.

I get the following error message everytime I try to make the change under Multiline Set Group Assignment: "cannot change from single line or threshold alert to group"

The only way to change it is to add another appearance of that DN and designate it as a "Multiline". As long as the DN has only 1 appearance it is a "Single Line". The system determines ththe status of the original Prime Line type.

Thanks for the tip; what's the maximum number of appearances a single DN can have?

I think that would be 32 counting the original DN. You would find that in Multiline Broadcast Groups.
