Has anyone ever set up Unified Messaging on a Toshiba iES16 XPE? If so, what do I need to make sure of before it will work. I know all of the obvious like enabling UM in the mailbox, email address, etc.

Any help would be much appreciated!

From memory its pretty simple on the IES and you have covered the obvious already. The main problems I have found were with permissions on the clients LAN to allow the IES access to perform the necessary tasks. A bit of co-op required with their IT support I'm afraid...
Not mentioned above is licensing, however if you are able to enable UM in the mailbox, you have licenses available. Its usually just as simple as enabling UM, enter the users email, and enter the address of the SMTP server, I usually use just the IP address for this if the server is in-house. As Otternose mentioned, the server needs to be able to accept these messages from the iES.
Even without providing an SMTP server the Stratagy will attempt to send the message directly to the domain of the email address provided in the UM tab of the user. A growing problem is when companies are using cloud based exchange servers. Often the message delivery is rejected as spam. Some IT guys will provide an account on these servers, but the Stratagy can not do athentication.

In some cases I have used the SMTP address of the ISP for that company. If you are on that network, sometimes they will allow the use of their SMTP without athentication.

Also, by default there is no domain setup for the STratagy. This will cause the message to be rejected by some SMTP servers. I actually found out it is sometimes better to use a domain that is not part if the company, something made up like stratagyvoicemail.com. If you use the same domain as the company, the SMTP server will check the name (Usually call "Unknown" or a caller ID) and reject the message due to that user not being part of their network.

Best start is to fill in the email address in UM tab, SMTP if you have one. Do a test. If no email is recieved,then check logs or run a trace.
Learning how to use the 'Trace' is the most critical thing I've found for troubleshooting UM problems.

You can set the filter to filter out just the UM related messages, or color code them.
