After I did the test that Phonemeister suggested I found the baud rate mismatch that exactly Breed suspected.
The Backup system PIOU1 has max baud rate 1200. I got the right output on the hyperterminal after I corrected the jumper for tty. Thanks Phonemeister.
The MAIN system uses 2400 baud rate as I could not get the output with 1200 setting on the hyperterminal. I got the output on hyperterminal with 2400.
I probably have to buy RSIU card to get 2400 baud rate unless there is any other card with some more useful features.

I'm not sure if you need the RSIU since you have a working PIOU. Just set the Stratagy for 1200 baud for SMDI.

Thanks so much. Even though I guessed that it would work by changing Stratagy to 1200, I first hesitated not to take any risk as it was working fine with the Main dk280 unit. Your reply gave me confidence and I changed the baud rate to 1200 for backup unit, it worked fine. Then I changed it back to 2400 for the Main system. I don't think that RSIU1 has any jumper to chaage the baud rate to 1200 that way Main and Backup unit will be at 1200 and we don't need to change the Stratagy setting every time. Other option is that I do have a PIOUS card on the main unit that has a TTY port, may be I can switch the cable from RSIU to PIOUS. Not sure if there is any disadvantage. Thanks so much again. I really appreciate all your help.

MSG LED: On Backup system somehow I could not get MSG button LED lit/flash even though there was a new message left for that intercom Ext/mailbox. It works fine on the Main system. On the Stratagy end only change I made was to set baud rate to 1200 for backup unit that uses PIOU card. I think I did set all the programs required for VM on Backup 280 unless I missed something. Is there any thing that I should verify to make MSG led flash, also to enable MSG key to retrieve message. Thanks in adavnce.

Closing this issue as I was able to resolve all except MSG Led with your help. Thanks a lot phonemeister, breed, Old blond hippity hopping Bunnie for your time. For MSG Led, I will add a new topic with details about the setup and programs I ran. This is a great forum. Thanks again.

Just want to update for future visitors if they face similar issue. I was able to resolve MSG Led issue, the problem was in Program 3, RCTU was set to 91 instead of 92 with RRCS option. It seems RRCS option was required for MSG light to flash. Then I ran into another issue when I pressed MSG button to retrieve the message I was getting message from voice mail some thing like invalid access code even though on the display I could see that it was forwarded to the voicemail extension. I remembered Phonemeister advice to remove #656 because voicemail was setup as SMDI. But it did not click to me to remove #657 also. Anyway after spending some time when I removed #657 everything worked fine. Thank you all.
