I have an Analog Port Adapter connected to a DTU-16D phone. The port is connected to a PC. So when a phone number gets generated from the PC the phone picks the 4th line and dials out.
How can I make it pick the 1st line instead?

Outbound call hunting usually goes from last to first to avoid collisions. Not sure if you can change that on a Elite 48. You could just change the order they are connected to your system. But the collision thing might still be an issue. I would have the local telco set the same caller ID for all your lines.
