Keylink DX, Comdial / Vertical DX-80. I want to have routing box 801(night) transfer from (both)CO lines DIRECTLY to voicemail box 101, without transferring to the switch / ringing the phone extension. I have done this before, and know that it is changing one *,* entry in trans.txt, but I cannot find documentation on configuration of the trans.txt anywhere. I also do not want it to play any of the system messages like "the person at this extension is not available, to leave a message, press 1", etc.

Please be a bit more specific here on what you want to do. Remember changing the trans.txt table is global and not dependant on day/night service. If you are trying to transfer calls to certain extensions during night service, you will do that in box 801. If you don't want the system message, record a custom greeting to override it.

Change the transfer type on mailbox 101 to No Transfer. If that's not an option (options were limited on some of the DX80 voicemails), change the mailbox status.

Regarding the second request, check your installation manual. I'm assuming you only want this behavior on the one mailbox. If so, there's an entry you can make at the mailbox level. Keep in mind that part of the "generic greeting" is because no voicemail greeting has been recorded by the mailbox owner, but the second part, "Please speak after the tone...", can be squelched and instead go right to the beep right after the greeting.
