Hello,
I am trying to connect a 85B1 transformer for an 82B and 82A connecting block for a 4A speakerphone setup.
The transformer was connected to the block via yellow/black wire.
There are two terminals on the transformer, labelled 3 and 4, and 2 AC terminals in the 82A/B connecting block.
Does polarity matter in this case?

No, polarity does not matter. It is AC.
