I've been asked to connect a MOH unit to the Nitsuko DX2E-24M, does anyone now if i need additional hardware i.e Misc card for connection and, whether programming is needed or an external jumper switch is used.

Yes programming will probably be needed, no additional hardware is required. MOH is a pair off the CPU
