The Day 2 transfer mode on the DSS console disables our backline numbers from picking up. I am using the DKI Admin program to configure it. They pickup fine on Day1 and Night transfer. How do I enable the backline numbers to pickup?

I think I was able to load the right settings ok copying settings from Night Transfer mode I uploaded and saved them. I forgot one thing though - do I need to reboot to get settings to take?

Ringing programs do not need a reset to take effect. You said you were using DKi admin to administer the database - it will prompt you if a reboot is needed.
