I am about to upgrade my Norstar CICS from 6.0 to 6.1 to enable a T7316e with a T24 KIM. Will this be plug and play if I just change the cartridge, or will I have to re-program? Thanks for any guidance!

It's plug and play. Be sure you are using at least 6.1MR. 6.1 will change your line numbers of the 2nd line card, but the upgrade will take care that for you.

Does this also apply to a Modular ICS upgrading from a 6.0 to a 6.1?
